Description

RPMI 1640 is suitable for a wide range of cells. RPMI 1640 Medium is commonly supplemented with 10% Fetal Bovine Serum (FBS). Although, RPMI-1640 has been shown to support the serum-free growth of a number of cells.

  • With L-Glutamine
  • With 2.0 g/L sodium bicarbonate
  • Without HEPES buffer

Applications

RPMI-1640 medium was developed by Moore et al., at Roswell Park Memorial Institute, hence the acronym RPMI. The formulation is based on the RPMI-1630 series of media utilizing a bicarbonate buffering system and alterations in the amounts of amino acids and vitamins. RPMI-1640 medium has been used for the culture of human normal and neoplastic leukocytes. RPMI-1640 when properly supplemented, has demonstrated wide applicability for supporting the growth of many types of cell cultures.

RPMI 1640 Medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 Medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 Medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). Although, RPMI-1640 has been shown to support the serum-free growth of a number of cells.

Common applications of RPMI-1640 include fusion protocols and growth of hybrid cells, the culture of normal human leukocytes in monolayer and suspension, and support of HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinoma growth (to name a few).

RPMI-1640 and DMEM are both commonly used media for mammalian cell culture. What are the differences between RPMI-1640 and DMEM?

  • • RPMI-1640 contains glutathione, a reducing agent, and high concentrations of vitamins not found in DMEM
  • • DMEM is used in culturing adherent cells
  • • RPMI is used for culturing cells in suspension
